Exocyst Subunits Interactions with Membrane Lipids Specific Targeting to Endomembranes

Charles University

Octameric complex exocyst was discovered, and is currently studied, in the context of exocytosis regulated by small Rab and Rho GTPases. We have described exocyst in plants and proved that it has functions in the secretory pathway and cell morphogenesis including cytokinesis, root and root hair development, seed coat development, polar auxin transport and plant defence against pathogens. Analysing specific exocyst subunit LOF mutant in Arabidopsis we have recently discovered that for some exocyst subunits there is also a function in a specific membrane transport pathway to the vacuole.

Evolution of Brain Size and Number of Neural and Glial Cells in Birds

Charles University

In this project, a postgraduate student will estimate, utilizing the isotropic fractionator method combined with flow cytometry cell counting, the total number of glial and neuronal cells and glia/neuron ratio in whole brains and parts thereof in four model groups of birds: the gallinaceous and passerine birds, owls and parrots.

Effect of Gregarines on Leishmania Infection in Sand Flies.

Charles University

Within the proposed PhD project we would like to test the coinfections of the gregarines and L. tropica and study the mechanism of immune response of sand flies to gregarines and Leishmania. We focus on expression of peptides involved in the immune response during Leishmania infection (e.g. defensin, cecropin, TEP and serpin); presences of which we described previously (Boulanger et al., 2004, Dostalova et al., 2011).

Characterization of Unusual RNA Polymerase Encoded By Yeast Linear Plasmids

Charles University

The project aims to: verify in silico predicted function of ORF6 and ORF7 encoded by pGKL2, as a putative RNA polymerase and its putative subunit, respectively, using various experimental approaches including heterologous protein production, testing for RNA polymerase activities and mutation studies of the RNA polymerase active site.

Evolutionary Significance of Free-living Metamonads

Charles University

The aim of the project is to investigate the phylogenetic position, morphology and cell structure of the newly discovered free-living metamonad lineages. The research will be supplemented by an analysis of transcriptomes of two novel metamonads. The results will significantly contribute to the knowledge about the early evolution and origin of parasitism in metamonads.

Evolution of Sex Determination and Sex Chromosomes in Squamate Reptiles

Charles University

Reptiles possess variability in sex determining systems ranging from environmental sex determination, where sex chromosomes are not present, to well-differentiated XX/XY or ZZ/ZW sex chromosomes. This variability is particularly substantial in squamate reptiles, the group encompassing lizards and snakes.

Vivo Study of Migration and Differential Potential of Xenopus Tropicalis Testicular Stem Cells

Charles University

In cooperation with the University of South Bohemia in Ceské Budejovice (Faculty of Fishery) interspecies TSC transplantations (between amphibians and fish) will reveal the rate of evolutionary conservation in migration behavior and ability to respond to xenogenous differentiation signals.
